Abstract: This study describes the development of fermentation technology for the secretory production of recombinant, human chymotrypsinogen B using the methylotrophic yeast Pichia pastoris in an integrated approach of genetic engineering, fermentation and downstream processing. Three different fermentation approaches have been developed, each respecting the demands of an industrial production process in a different way. In a fed-batch process using methanol as the sole source of carbon and energy during the production, a space-time-yield of 5.4 mg$\cdot$1$^{-1}$ $\cdot$h$^{-1}$, combined with a high product titer of 480 mg$\cdot$1$^{-1}$ and biomass concentration of 150 g$\cdot$1$^{-1}$ cdw has been achieved. Avoiding the high oxygen demand of the latter high-cell-density process, two different continuous approaches have been investigated on, yielding space-time-yields of 25 mg$\cdot$1$^{-1}$ $\cdot$h$^{-1}$, using methanol as the sole substrate and 10.3 mg$\cdot$1$^{-1}$ $\cdot$h$^{-1}$ in a simplified mixed-feed approach using glucose and methanol, respectively.
